Here's the short version. CJC-1295 and ipamorelin have short human pharmacology studies, and ipamorelin was also studied after bowel surgery. These records do not establish a body-composition, sleep or energy timetable [6].
The human studies measured hormones for days, not results for months
The selected human CJC-1295 reports are short hormone studies in healthy adults. FDA summarizes 63 healthy adults and says 73% received one injection. It explicitly identifies the eleven-man Sackmann-Sala analysis as a subset of the Ionescu study. The reviewed summary does not resolve a unique participant total across all publications; publication counts should not be added as independent treatment cohorts [6].
| Time after a dose | What changed | Peptide | Study |
|---|---|---|---|
| About 40 minutes | One growth-hormone pulse peaked, then fell to almost nothing | Ipamorelin, by vein | Gobburu 1999 [4] |
| About 2 hours | Half the drug cleared from the blood | Ipamorelin, by vein | Gobburu 1999 [4] |
| 6 days or more | Growth hormone 2 to 10 times higher | CJC-1295 (DAC form) | Teichman 2006 [1] |
| 1 week | Mean growth hormone +46%, IGF-1 +45%; pulse rhythm unchanged | CJC-1295 (DAC form) | Ionescu 2006 [2] |
| 9 to 11 days | IGF-1 1.5 to 3 times higher | CJC-1295 (DAC form) | Teichman 2006 [1] |
| Up to 28 days | IGF-1 still above baseline after repeated doses | CJC-1295 (DAC form) | Teichman 2006 [1] |
| Weeks to months | Muscle, fat, sleep, energy or aging | Either, or both | Not established in cited studies |
A rise in IGF-1 is a lab number, not a result you'd notice. Growth hormone and IGF-1 going up says the peptide reached its target. It does not establish an improvement in strength, function, sleep or health.
| Study | Peptide | Who, how many | Design | What it measured | Result |
|---|---|---|---|---|---|
| Teichman 2006 [1] | CJC-1295 (DAC) | Healthy adults 21 to 61 | Two placebo-controlled trials, 28 and 49 days | Growth hormone, IGF-1 | Both rose for days; half-life 5.8 to 8.1 days |
| Ionescu 2006 [2] | CJC-1295 (DAC) | Healthy men 20 to 40 | Before and after one dose | Growth-hormone pulses | Trough level 7.5 times higher; rhythm kept |
| Sackmann-Sala 2009 [3] | CJC-1295 | 11 healthy young men | Before and after one dose | Blood proteins | A few protein markers shifted |
| Gobburu 1999 [4] | Ipamorelin, by vein | Healthy men, 8 per dose level | Dose-escalation | Drug levels, growth-hormone pulse | One short pulse; half-life about 2 hours |
| Beck 2014 [5] | Ipamorelin, by vein | 114 analyzed after bowel surgery | Phase 2, placebo-controlled | Time to first solid meal | 25.3 vs 32.6 hours, not significant |
| Not established in cited studies | The combination | Not established | Not established | Not established | Unknown |
A hormone timeline does not establish a visible-results timeline
The cited studies do not establish claims such as energy in week one or body changes by weeks8to12 for this combination. Ask for the supporting trial and its actual measured outcome.
If a page gives you a timeline, ask what it's based on. A hormone rise within days is real. A visible change at a set week is a guess.
"CJC-1295" on a label may not be the molecule that was studied
The studies above used the DAC form, a version built to bind to blood proteins so it lasts for days. A version made without DAC is often sold under the same name. FDA's reviewers flagged the naming itself as a safety risk [6].
There appear to be inconsistent naming conventions associated with CJC-1295-related BDSs. This represents a safety risk for patients as they may be dosed with a different BDS than the physician ordered.[6]
In plain terms: the hormone timings in the table describe one specific molecule. A product without DAC wouldn't be expected to last days, and it hasn't been studied the same way. Ask your prescriber which molecule anyone is talking about.
FDA's advisers voted against both peptides in 2024
FDA's compounding advisory committee voted against adding ipamorelin in October 2024 and all five CJC-1295 forms in December 2024 to the 503A list of substances pharmacies may compound.
| Date | What happened |
|---|---|
| September 29, 2023 | FDA places ipamorelin acetate in Category 2 of its 503B list, for outsourcing facilities, where it remains [10] |
| October 29, 2024 | Advisory committee votes 0 yes, 12 no, 1 abstain on adding ipamorelin, in both forms, to the 503A list [8] |
| December 4, 2024 | Advisory committee votes against all five CJC-1295 forms: 0 to 13 for four, 1 to 12 for the acetate [7] |
| May 14, 2026 | FDA's 503A list update shows neither peptide in any category [11] |
Neither has ever been FDA-approved. Both are also on the World Anti-Doping Agency's 2026 list, banned at all times [12]. The full legal picture is on the legal status page for ipamorelin and CJC-1295.
FDA's reviewers list real side effects for both
FDA's briefings describe injection-site reactions, flushing and headache with CJC-1295. For ipamorelin, its one phase 2 trial saw more high blood sugar than placebo.
- CJC-1295, injection site: irritation, redness or itching in about 70% in one study and in everyone on the drug in the other [6].
- CJC-1295, headache: 63% on the drug versus 14% on placebo in the first study.
- CJC-1295, flushing: in 40% at the low dose and everyone at the high dose, usually within 30 minutes. FDA also notes a faster heart rate [10].
- Ipamorelin: high blood sugar at discharge in 14.3% versus 8.6% on placebo, and low potassium in 12.5% versus 3.4% [9].
If growth-hormone support was the goal, sermorelin is the legal relative
Sermorelin, a growth-hormone-releasing hormone analog, is available by prescription as a compounded drug. Compounded drugs aren't FDA-approved. The route is in how to get sermorelin, and its own timeline is in how long sermorelin takes to work.
